Tolerance is the whole craft
Real photos rarely contain one flat color — a "white" wall is a hundred near-whites in shadow and light. Low tolerance changes only near-exact matches; raising it widens the net (with a soft edge so the change blends). Start around 40, apply, and adjust — every Apply starts fresh from your original, so experimenting is free. For changing a photo background cleanly when the subject has complex edges (hair, fur), a dedicated AI background remover is honestly the better tool; flat and simple images are where color replacement shines.

Lower the tolerance, and pick the source color from the exact spot you mean (zoom by using Pick from image on that area).
Can it make a color transparent?
Not in this tool — replacement is color-to-color. PNG transparency work belongs to background-removal tools.
